RNA Production by Bacillus mesentricus niger

Twenty-four strains of various Bacilli were compared as to the productivity of extracellular RNA. Some strains, especially B. mesentricus niger IFO 3214,produced extracellularly considerable amounts of RNA on maltose medium. Strain no. 6021 derived from B. mesentricus niger IFO 3214 on NTG treatment produced 2.5 mg per ml of RNA and was sensitive to SDS. Using derivative no. 6021,the culture conditions of this RNA-excretive bacterium were investigated using 8% maltose as a carbon source. The productivity of RNA was greatly dependent on the initial pH of the growth medium, and the amount of RNA produced reached 4.2 mg per ml of the culture. Furthermore, a high level of aeration resulted in good growth and a high yield of RNA. The process of leakage of intracellular RNA from the intact cells occurred without any lysis. When strain no. 6021 was cultured on maltose medium at initial pH 8.0 and the working volume of the medium was 25 ml, the RNA yield was 5 mg per ml of the culture. This amount of extracellular RNA of strain no. 6021 was an increase of about 4 times as compared with that of the parental strain, IFO 3214.
