Raw Starch Adsorption, Elution and Digestion Behaviour of Glucoamylase of Rhizopus niveus

A Rhizopus niveus glucoamylase preparation consisted of five forms which were separated by preparative isoelectric focusing. All of them were strong in debranching activity, raw starch adsorption and digestion. Among them, two major forms termed glucoamylase C and glucoamylase D having specific activities of 8.55 and 9.23 units/mg protein, debranching activities of 0.46 and 0.40,isoelectric points of 8.45 and 9.1,carbohydrate contents of 14.9 and 12.7%, and hydrolysis limits of boiled soluble starch of 62 and 67%, respectively, were used in this investigation. Both amylases were completely adsorbed onto raw starch. Optimum adsorption of glucoamylase C and glucoamylase D onto raw starch occurred at pH 3.5-7.0 and 5.0-5.5,respectively, and the pH was not so important for adsorption. The adsorption was affected by temperature. Amylase inhibitor from Streptomyces sp. could not prevent the adsorption of these amylases onto raw starch although the amylases could not digest raw starch in the presence of it. Borate in alkaline conditions eluted about 25% of both the adsorbed enzymes from raw starch. The optimum pHs for boiled soluble starch hydrolysis by glucoamylase C and glucoamylase D were 5.5 and 5.0 whereas those of raw starch digestion were 4.0-6.0 and 4.5-6.0,respectively.
