Interaction of Lactobacillus plantarum Phosphoglycerate Kinase with Cibacron Blue F3GA

Phosphoglycerate kinase (EG 2.7.2.3) of Lactobacillus plantarum AHU 1047,adsorbed to a blue dextran-Sepharose 4B column, was eluted quantitatively with 1 mM pyrophosphate. Pyrophosphate inhibited the phosphoglycerate kinase competitively with respect to both 3-phosphoglycerate and ATP. Further inhibition studies suggested that two molecules of pyrophosphate bound to one molecule of the kinase. The difference spectrum of Cibacron Blue F3GA in the presence of the kinase was similar to that in the presence of KCl. The difference extinction coefficient at 680 nm was decreased by the addition of pyrophosphate, 3-phosphoglycerate, or ATP. The kinase was inactivated by treatment with pyridoxal phosphate. This inactivation was prevented by pyrophosphate, 3-phosphoglycerate, and ATP, as well as Cibacron Blue F3GA.
