Fluorescent X-ray imaging with synchrotron radiation for the evaluation of probes for biomedical functions : A pilot study on 4-bromo-DL-phenylalanine for cancer detection in mice

A pilot study for a fluorescent X-ray imaging system was performed at beam line BL-15B1 of the Photon Factory storage ring (2.5 GeV) at the High Energy Accelerator Research Organization in Tsukuba, Japan. We used a phantom to test the minimum detectability and quantitativeness of bromine in this system. A slice of tumor-bearing mouse (body weight, 30g) was visualized after the administration of 0.1mL of 1.0mM bromine-labeled compound, 4-bromo-DL-phenylalanine (Br-Phe). A linear relationship was observed between the concentration of bromine and the fluorescent X-ray count in the phantom experiment. The minimum detectability of bromine in this system was found to be 107 μM. The two-dimensional distribution of bromine was depicted in the section of tumor-bearing mouse. In the tumor region, the bromine concentration was estimated to be approximately 500 μM. Considering the minimum detectability of bromine in this system, this method enables tumor-imaging using non-radioactive Br-Phe.
