Phosphoglycerate kinases (EC 2.7.2.3) of sevral lactic acid bacteria were adsorbed to blue dextran-Sepharose 4B and eluted with 1 M KCl. At 1 mM, 3-phosphoglycerate, ATP, ADP, GTP, GDP, NADP and NADPH could elute phosphoglycerate kinase of Lactobacillus plantarum AHU 1047 from blue dextran-Sepharose with good yeilds, but AMP, GMP, NAD and NADH were less effective in eluting the kinase. It should be noted that the presence of phosphate at the 2'-position of the ribose moiety of pyridine nucleotide had a remarkable effect in eluting phosphoglycerate kinase form blue dextran-Sepharose. By a single step purificaiton using a linear gradient of 3-phosphoglycerate, between 0 and 1 mM, the kinase of L. plantarum AHU 1047 was purified to be electrophoretically homogeneous. Cibacron blue F3GA inhibited the enzyme competitively with respect to 3-phosphoglycerate and noncompetitively with respect to ATP.
