Affinity Chromatography of Candida Pyruvate Kinase on Bule Dextran-Sepharose

Pyruvate kinase(EC 2.7.1.40)of Candida guilliermondii AHU 4101 was absorbed to a blue dextran-Sepharose 4B and eluted with 1 M KCl. At 30 mM concentrations, ATP, ADP, GTP, CTP, NADP(H), phosphoenolpyruvate and fructose 1,6-bisphosphate eluted the kinase from the blue dextran-Sepharose column, but AMP, NAD(H), pyruvate and fructose 6-phosphate did not. By a two-step purification procedure using blue dextran-Sepharose affinity chromatography followed by gel filtration on Sephadex G-150,pyruvate kinase of C. guilliermondii was purified approximately 18-fold with a yield of 29%. The purified enzyme appeared to be homegeneous on 5% polyacrylamide gel disc electrophoresis. In the presence of 0.5 mM fructose, 1,6-bisphosphate, blue dextran inhibited the kinase competitively with respect to both phoshoenolpyruvate and ADP.
