Effect of conditioning stimulation of the fornix on the response of thalamic nociceptive neurons to tooth pulp stimulation

Using cats anesthetized with urethane and chloralose, we studied the effect of conditioning stimuli applied to the fornix(Fx)on the response of nociceptive neurons with tooth pulp(TP)afferent input in the nucleus ventralis posteromedialis(VPM)and in the intralaminar nuclei(IL)of the thalamus. In half of the IL neurons tested, Fx conditioning stimulation at an intensity of less than 1 mA inhibited responses to TP stimulation and to tract stimulation. The inhibition of response of IL nociceptive neurons was partially antagonized by intravenous administration of naloxone(1 mg/kg). However, Fx conditioning stimulation did not inhibit the response to TP stimulation or to tract stimulation in any of the VPM neurons. These results suggest that inhibition arising from the Fx acts mainly at the thalamic level on the medial pain pathway and that this inhibition is mediated in part by opioid receptors.
