Expression of MMP-2 and HB-EGF in keratocystic odontogenic tumors

Some odontogenic cysts have been found to have special biological features that make them distinct from other lesions. This study was conducted to detect the expression of MMP-2 and HB-EGF in keratocystic odontogenic tumors (KCOTs) and to examine the possible predictive value of these markers. Paraffin-embedded tissue specimens of 11 KCOTs were immunohistochemically examined using antibody against MMP-2. Expression of the EGF family mRNA in KCOT was analyzed by real-time RT-PCR. Fathermore, expression of HB-EGF mRNA and protein were examined in KCOT. Nine of the 11 KCOT cases were immunohistochemically reactivity for MMP-2. The immunopositive reaction to MMP-2 was strongly expressed in the cytoplasm of the prickle cells. The expression levels of HB-EGF were high in KCOT. In addition, although high expression levels were detected for amphiregulin (AREG), epigen (EPGN), epiregulin (EREG) and TGF-α mRNA, betacellulin (BTC) and EGF mRNA were not highly expressed. There was high expression of the amount of HB-EGF protein when compared with the primordial cyst. These findings suggest that the aggressive behavior of KCOTs might be explained by the expression of MMP-2 and HB-EGF, which may be valuable markers for the prediction of the biologic behavior of KCOT.
