Study on diurnal variation of noradrenaline release in three brain regions of rats.

Diurnal variations of noradrenaline (NA) release in three brain regions were investigated by measuring 3-methoxy-4-hydroxyphenylethyleneglycol sulfate (MHPG-SO<SUB>4</SUB>) level. Male Wistar rats were kept in a temperature controlled room (24±1°C) with lights on from 7:00 A. M. to 7:00 P. M. during the month of July, and killed at twelve different times of the day. NA and MHPG-SO<SUB>4</SUB> levels in the pons+medulla oblongata, hypothalamus and cerebral cortex, and plasma corticosterone levels were assayed fluorometrically. The rats showed normal diurnal rhythm in plasma corticosterone levels, i.e., highest at the end of a light period and lowest at the end of a dark period. Levels of NA in the pons+medulla oblongata and hypothalamus, and of MHPG-SO<SUB>4</SUB> in the three regions, varied during the course of a day. Statistically significant differences were observed between maximum and minimum values of these levels. Changes in MHPG-SO<SUB>4</SUB> levels, however, were not rhythmic. These results suggest that NA release does not demonstrate a diurnal rhythm in the pons+medulla oblongata, hypothalamus or cerebral cortex.
