ハムスター卵円形細胞の肝吸虫感染による分化

• 卵円形細胞ovall cellは肝障害後に現れ, 肝細胞と胆管細胞の母細胞と考えられている.ジエチルニトロサミンと2-アセチルアミノフルオレンを投与して肝吸虫(Clonorchis sinensis; CS)を感染させたハムスターから分離した卵円形細胞を2週間培養し, その分化と可塑性について電顕と免疫染色で調べることによって評価した.CSを感染させていない細胞の2週間培養で, グリコーゲン顆粒とぺルオキシソームがみられた.免疫組織化学的に培養1週間後からのサイトケラチン19に対する明瞭な反応が増加し, アルファフェト蛋白に対しては徐々に減少した.このことはCSに感染していないハムスターより分離した卵円形細胞は肝細胞系に分化したことを意味した.しかし, CS感染グループからの培養細胞は, 多くの粗面小胞体を有し, CS感染細胞からのものと相対的に逆の反応を示した.このことはCS感染からの分離細胞はCSに最初に刺激されて胆管系の方向へ分化したことを意味した.この研究の結果, 卵円形細胞は実際には肝細胞と胆管細胞の両極性の母細胞で, 最初に刺激する要素によってどちらかの方向へ分化可能であることを示唆している.
