ナチ毒(BV)の注射は, 持続性の疼痛と痛覚過敏をもたらす一方、BVが抗炎症作用と抗侵害受容効果を及ぼすという証拠がある.BVは, この目的で伝統的に東洋医学で痛みの緩和や慢性の炎症性疾患, 例えば慢性関節リューマチの治療に用いられてきた.本研究は, BVが正常状態下で急性の侵害受容反応をもたらすが, 局所炎症反応において強力な抗侵害受容効果と抗炎症作用を示すという仮設を検討するために計画した.ラットへのカラゲナン(CR)投与による急性の足浮腫と温痛覚過敏に対するBV前処理の効果を評価した.更に, 末梢の炎症による脊髄でのFos発現を定量的に解析した.正常動物では, 後肢皮下へのBV投与が明瞭な侵害受容性行動や痛覚過敏を伴うこと無しに, 僅かながら脊髄のFos発現を増加させた.一方, BV(0.8mg/kg)をCR投与30分前に投与すると, CRによって誘起される足浮腫と温痛覚過敏を抑制した.この時, 足の体積の百分率変化と脊髄のFos発現ニューロンの数の間に正の相関関係が認められた.これらの結果は, BVの前処理がCRによって誘起される炎症性の痛みに対して抗侵害受容効果と抗炎症作用を表すことを示している.更にこれらの結果は, BV投与が慢性的な炎症性疾患に伴う痛みと浮腫に対する有効な処置であることを示唆している.
