Purification and Some Properties of Plant Serine Acetyltransferase

Serine acetyltransferase (SATase) that catalyzes the conversion of L-serine to O-acetyl-L-serine (OAS) in the presence of acetyl-CoA was highly purified from rape leaf extract, using a coupled assay system in which OAS is converted to cysteine by enzymatic action of exogenous cysteine synthase. Through purification procedures including heat treatment, ammonium sulfate fractionation and successive column chromatographies on DEAE-Toypearl, Blue-Toyopearl and Toyopearl HW-55, the specific activity was raised to about 4,800-fold over that in the crude extract. The molecular weight of rape enzyme was estimated to be about 350,000 by gel filtration column chromatography. The cysteine-forming activity of the final preparation was completely dependent on L-serine, acetyl-CoA and sulfide. However, this preparation had low activity for L-cysteine synthesis from L-serine even in the absence of exogenous cysteine synthase, suggesting that plant SATase exists as a high-molecular weight enzyme complexed with cysteine synthase.
