The biosynthesis of sepiapterin and biopterin in rats was studied. When [U-^<14>C] GTP was injected into rats, radioactivity was incorporated into these pterins isolated from urine, whereas incorporation from [8-^<14>C] GTP was scarcely observed. The incorporation into both pterins was greatly enhanced by chclohydrolase-treatment of [U-^<14>C] GTP. The enhancement was especially striking in the case of sepiapterin. These results suggest the biosynthetic pathway of sepiapterin from GTP by way of dihydroneopterin thiphosphate in rats.
