Isolation of a polysaccharide from the inner cell wall, intine, of pollen of Cryptomeria japonica

Isolation and solubilization of the intine of pollen grains of Cryptomeria japonica were attempted. The intine was separated effectively from other cell fragments by passing it through a column of glass beads and dissolving it in an EDTA solution. The extract was fractionated by column chromatography on DEAE-cellulose and gel-filtration through Sephadex G-200, and a polysaccharide component was obtained. This was found to consist of galactose, rhamnose, arabinose, xylose and uronic acid.
