Rapid Transformation of Baciilus subtilis Using KCl-Treatment

An optimal simple and quick method for transforming Bacillus subtilis by plasmid DNA was developed. Cells exposed to 410 mM KCl solution were transformed with plasmid DNA in the presence of 35% polyethylene glycol. All steps before the gene expression stage were completed within an hour, and several thousand transformants were obtained from one μg of plasmid DNA. The activation of autolysin(s) of the cells is believed to be involved in the mechanisms of this transformation.
