Analysis of cytokines using a flow cytometer on lymphocytes obtained from human periodontitis gingivae by mechanical dispersion

Gingival tissues excised from patients with periodontitis were used for collecting mono-nuclear cells by mechanical dispersion. Collected cells were analyzed by flow cytometry to determine the balance of the ratio between Th1 and Th2 cells. Isolated cells from the peripheral blood of periodontitis patients (PPB), peripheral blood of healthy volunteers with healthy gingiva without systemic disease (HPB), and inflammatory periodontal tissues (IPT) were incubated with anti-IFN-γ and IL-4 monoclonal antibodies using the FASTimmune Cytokine System, and analyzed by flow cytometry. Mononuclear cells from IPT showed diminished IFN-γ compared to cells from both PPB and HPB. In contrast, IL-4 was significantly greater than that obtained from either PPB or HPB. These results suggest that cells producing IL-4 predominate in periodontally-involved gingivae. IL-4 produced by Th2 cells activates B cells, which then differentiate into plasma cells. This is followed by a cell population as found in IPT that may give rise to an abundance of non-protective antibodies that form harmful antigen-antibody complexes that induce the local destruction of periodontal tissue.
