Confocal laser scanning microscopic observation of the alignment of dentinal tubules branching from secondary canals in human teeth

Although dentinal tubules radiate from the root canal at right angles, it is not known how they branch from secondary canals. In order to elucidate the morphology of the secondary canals and the alignment of dentinal tubules branching from secondary canals, we observed the apical roots of molars in human teeth. Thirty-three adult human maxillary and mandibular molars were chosen for this experiment. Two types of specimens were made : transparent decalcified teeth with root canals permeated by India ink (transparent specimens) and undecalcified teeth with root canals permeated with alizarin red (alizarin red specimens). Both specimens were examined macroscopically or with a magnifying glass and observed with confocal laser scanning microscopy (CLSM). The alizarin red specimens were investigated in more detail than the transparent specimens using CLSM. The dentinal tubules originated at the cementodentinal junction, ran parallel to the secondary canals, and terminated at the root canal. At the branching where the secondary canals entered the root canals, the secondary canals formed a functional joining by dentinal tubules of different lengths.
