A simple and rapid assay for collagenase activity using fluorescence-labeled substrate.

A new assay method was developed for bacterial and animal collagenase activity, using fluorescence-labeled soluble collagen as a substrate. The enzyme reaction was carried out at 35°C in an incubation medium containing the substrate and 0.25 M glucose and the intensity of fluorescence emitted by the digested product was measured on a 50% dioxane extract of the mixture. Linear relationships were observed between fluorescence intensity and both enzyme activity and incubation time. It was confirmed that the method was a simple and rapid method for the assay of collagenase activity with high precision.
