Inhibition of Cytokinin-Induced Adventitious Bud Initiation in Torenia Stem Segments by Inhibitors of Serine Proteases

Application of di-isopropyl fluorophosphate (DFP), a highly sensitive inhibitor for serine enzymes, strongly inhibited cytokinin-induced adventitious bud initiation in Torenia stem segments cultured in vitro. The inhibitory effect was not evident when DFP was applied after 3 days of culture. Amount of DFP-binding proteins remarkably increased in superficial tissues of explants cultured for 3 and 4 days on a medium containing benzyladenine. At least 14 kinds of DFP-binding polypeptides were detected by SDS-polyacrylamide gel electrophoresis and fluorography. DFP-binding to some of these polypeptides was inhibited by a prior treatment with phenylmethylsulfonyl fluoride and N-p-tosyl-L-lysine chloromethyl ketone. From these results, it was suggested that some serine proteases might be related with biochemical events occurring during the initial stage of adventitious bud differentiation in Torenia stem segments.
