Membranes containing protease were isolated from the cotyledons of germinating pea seeds by differential and two successive sucrose density gradient centrifugations. Membranes were recovered in the microsomal fraction, but could be clearly separated from membranes carrying antimycin A-insensitive NADH-cytochrome c reductase (EC 1.6.2.1), which seem to come from the endoplasmic reticulum. The density of protease-containing membranes was 1.135. Membranes contained a lower amount of phospholipid as compared with the endoplasmic reticulum. Electron microscopic observations also showed that the membranes were different from the smooth-surfaced endoplasmic reticulum.
