After intravenous administration of Indomethacin in a male albino rabbit, the plasma concentration-time course followed two-compartment model for all doses examined, 5,10,and 20 mg/kg. Although the model is linear, the pharmacokinetic constants showed a dose dependency, the two rate constants and volumes for distribution increasing as the dose increased. When administered intraduodenally, Indomethacin was absorbed to a smaller extent in 5 mg/kg dose than in the other two doses. These dose dependencies were ascribed to the saturable binding to the protein which brought about the increase of free Indomethacin. The absorption of Indomethacin from its suspension was rapid enough to show a pattern similar to that from a solution. Biliary excretion of 10-25% of the dose was found, but reabsorption was not recognized.
