PHT, a Transmissible Plasmid Responsible for Phthalate Utilization in Pseudomonas putida

A 200-kilobase pair (kbp) DNA plasmid, named PHT, was detected in a strain of Pseudomonas putida, NMH102-2,that utillized phthalate effectively through the 4,5-dihydroxyphthalate pathway. A mutant defective in phthalate utilization (Pht^-) was isolated by Tn5 mutagenesis of the strain. The DNA region tagged with Tn5 was cloned from the mutant and used as a probe to clone the corresponding intact DNA responsible for the Pht^+ trait. The essential region for the Pht^+ trait was within a 7-kbp EcoRI region and the Pht^- mutant had multiple copies of this DNA region, into some of which Tn5 was inserted. A 1.2-kbp fragment of the 7-bp EcoRI region hybridized with the 200-kbp PHT DNA and also with a similar 130-kbp plasmid found in another Pht^+ strain of P. putida isolated independently. The Pht^+ trait was transmitted from strain NMH102-2 to a Pht^- P. putida strain with a frequency of 2×10^<-6> per recipient cell by mixing recipient cells with excess donor cells. All the Pht^+ conjugants harbored a 200-kbp plasmid, while the original recipient cells did not. These observations indicate that the gene(s) responsible for the Pht^+ trait of these strains is on the plasmid.
