Enzymatic Hydrolysis of Sweet Potato for Energy-Saving Production of Ethanol

Enzymatic hydrolysis of sweet potato starch was carried out using the combined action of pectin depolymerase and glucoamylase. Two systems were tested, one without heating and the other with low-temperature heating (75-85℃ for 5 min). For hydrolysis, sweet potato mash was adjusted to pH 4.5 and macerated with 0.05% pectin depolymerase at 45℃ for 2 h or more. When the mash was subjected to low-temperature heating after maceration with 0.01% pectin depolymerase, 12% (w/v) ethanol was produced after 48-h fermentation by employing 0.05% glucoamylase for saccharification. Without heating, a similar ethanol yield was obtrained after 96-h fermentation with a larger amount of glucoamylase, 0.4%. In the hydrolysis system without heating, hydrolysis reached less than 30%, even with a glucoamylase concentration as high as 0.6%. However, in the presence of yeast cells, the sweet potato starch was almost completely hydrolyzed.
