An instrumented system of a koji making process was developed for a kinetic study on the solid-state cultivation of Aspergillus oryzae var. brunneus W03 on steamed rice.Estimation of the growth of the microorganism in the koji was successfully done by monitoring CO_2 evolution.The water activity of koji decreased according to the growth A. oryzae, mainly because of an increase in reducing sugar.Optimum conditions for growth were not always similar to those for enzyme production. An initial water content of 40% was suitable for growith while the protease activity was higher at 35% or less, and the optimum temperature was 38℃ for growth whereas the protease was produced more at 30℃. At the stationary phase of growth, α-amylase activity still increased while protease as well as saccarifying activites were constant.A supply of CO_2 at 2 to 5% stimulated the protease production.
