Metabolism of Aliphatic Nitriles in Fusarium solani

Fusarium solani MN 7030,which could utilize succinonitrile as a sole source of nitrogen, was isolated from a soil sample. This fungus utilized acetonitrile, propionitrile, n-butyronitrile, succinonitrile or glutaronitrile as a sole source of nitrogen. Adiponitrile or malononitrile did not support the growth of the strain.n-Butyronitrile and acetonitrile were degraded without accumulation of the corresponding amides or acids as intermediary metabolites. However, in the succinonitrile culture, succinamic acid (HOOC・CH_2・CH_2・CONH_2) was accumulated in the broth. Cell-free extract obtained from the n-butyronitrile- or succinonitrile-grown mycelia showed both nitrilase and amidase activities. Aliphatic mononitriles and dinitriles with 1 to 4 acyl carbons were favorably degraded. Degradation of n-butyronitrile proceeded stoichiometrically to form equimolar amounts of n-butyric acid and ammonia. pH optima of the nitrilase and amidase were 7.5.
