Intracellular levels of pyridine coenzymes were determined in some strains of yellow and black aspergilli. The total pyridine coenzyme content was found to range from 1.14 to 2.15 μmol per gram of dried cells except that A. oryzae contained a smaller amount of oxidized coenzymes. The ratio of NAD+NADP : NADH+NADPH was calculated to be 1.6 to 2.1. Tracer experiments showed that A. oryzae, A. flavus, A. niger, A. awamori and A. usamii synthesized NAD from nicotinic acid and the salvage pathway was considered to be involved in the synthesis of pyridine coenzymes. Nicotinic acid ribonucleoside (NAR) was detected both in the culture filtrates and mycelial extracts of all the strains tested. A high level of radioactivity was detected in the nicotinamide ribose diphosphate ribose (NAmRDPR) fraction of the culture filtrate of black aspergilli.
