Growth Characteristics of Marine Methanol-utilizing Bacteria : Studies on Marine Methanol-utilizing Bacteria(II)

The growth characteristics of marine methanol-utilizing bacteria were investigated for the production of biomass from methanol.Initial methanol concentration of 4-6%(v/v)in a medium inhibited the growth of these bacteria. NaCl concentration of 1-4% and the cultivation temperature of 30-37℃ were optimal for growth. The optimum pH for the growth was 6.9 to 7.1 for the medium prepared with fresh water and 6.7 to 6.9 for the medium with sea water. These bacteria required vitamin B_<12> for growth and showed linear growth in proportion of the concentration of vitamin B_<12> up to 10ng/l.Under the defined conditions, continuos culture experiments were conducted using selected bacteria in a medium prepared with sea water. For strain YK 4030,the the growth yield to methanol reached 51% at the dilution rate of 0.2 hr^<-1>. For strain YK 2030,growth yield of 51% and productivity of 5.3g/l・hr were obtained at the dilution rate of 0.5 hr^<-1>. Maximum specific growth rates of strain YK 4030 and strain YK 2030 were 0.5 and 0.73 respectively.The cell compositions and amino acid profiles of these bacteria were similar to those of terrestrial methanol-utilizing bacteria.
