Isolation of Methanol-assimilating Bacteria and Optimum Medium Composition

Methanol-assimilationg bacteria were isolated by a 2-stage continuous enrichment culture specially designed for this experiment. Medium was passed through a column packed with soil samples to wash out microorganisms from soil samples into the first of the two continuous enrichment culture vessels. Fifty strains which could assimilate either methanol only or both methanol and methylated amines but not methane were isolated. They were considered to belong to "the genus Methylomonas".Continuous culture of one of the isolates, Methylomonas sp. C-1012,was conducted in a basal medium containng proportional amounts of elemental components to the mean relative concentrations of the elements in bacterial cells. Depending on the surplus and deficit of each component in the basal medium at the steady state, the minimum medium composition to support normal growth of the cells was reconstituted.Continuous coltures in which nutrient concentration in the minumum medium was changed suddenly were carried out to determine the optimum medium composition for maximum growth yield with the complete consumption of the medium components under steady state in continuous culture. Additional supply of 1.0 g/l of KH_2PO_4 and 15 ppm of ZnSO_4 to the minimum medium gave the highest growth yield, 0.480 g・cell/g・methanol, with a cell mass concentration of 19.2 g/l and productivity of 5.68 g/l・hr.
