Isolation and Characterization of Marine Methanol-utilizing Bacteria : Studies on Marine Methanol-utilizing Bacteria(I)

In an attempt to utilize sea water for the production of a biomass from methanol, screening was conducted using a medium containing methanol and 3%(w/v)of NaCl, and 645 strains of methanol-utilizing bacteria were isolated.These isolates were obtained from sample of marine origin such as sea water, sea sand, sea mud, sea weed and so on, not from sample of terrestrial origin. The were Gram-negative rods, motile with a single polar flagellum and aerobic. Moreover, they required Na^+ ion and vitamin B_<12> for growth. The optimum range of Na^+ ion for growth was 1 to 6%(w/v)as NaCl. All of the isolates assimilated methanol and formaldehyde as sole carbon sources but not methane. Also, most of the isolates assimilated methylamiones and/or sructose.The cells of the isolates showed marked lysis when suspended in distilled water. This phenomenon is generally accepted to be one of the important properties of marine bacteria.On the basis of theis origins and properties, all of the isolates can be desighnated as marine methanol-utilizing bacteria.
