Culture conditions were investigated for the optimum production of L-glutamic acid from benzoate with Brevibacterium sp. No.6,one of the most productive strains. It was found that a biotin concentration of 3〜5 μg/l in a medium that is suboptimal for growth, pH control at 7.8,and a cultivation temperature of 29℃ were ideal as was an ample oxygen supply. Ammonium benzoate was the most favored as the substrate. It was added to the culture medium by intermittent feeding, in pararell with its consumption, to lessen its adverse effect on the cells at higher concentrations. Under these optimum conditions, L-glutamate accumulation reached about 80 g/l after about 40 hr culture. The yield was 85% by weight, based on the consumed benzoate. Some kinetic studies on this fermentation were also reported.
