Development of Dentin Regeneration Therapy : Expression of Type I Collagen and Alkaline Phosphatase Induced by CTGF in Human Cultured Dental Pulp

With the concept of minimal intervention (MI), there is an urgent necessity to develop new therapy of pulp capping and hard tissue regeneration. Recent studies have reported therapies using Ca(OH)_2 or/and adhesive resins or antibiotics mixture. In this study, we used connective tissue growth factor (CTGF) to develop a new method of biological pulp capping. To elucidate the ability of CTGF to induce tissue mineralization, we analyzed the expression of type I collagen and alkaline phosphatase, an indicator of tissue mineralization. Normal human pulp tissue was stimulated by CTGF in culture medium for several hours, and the expression of type I collagen and alkaline phosphatase were analyzed by immunohistochemical staining. Type I collagen was expressed markedly within 24 hours after CTGF stimulation and ALP was expressed 96 hours after stimulation. These results show that CTGF is extremely useful for the establishment of biological dentin regeneration therapy.
