Effects of ethanol on the maturation and secretion of hepatic secretory glycoproteins.

The effects of ethanol and acetaldehyde on the maturation and secretion of hepatic glycoproteins were analyzed using the pulse-chase labeling method and autoradiogram following two dimensional gel electrophoresis. A spot for transferrin was detected at 77, 000 in molecular weight and 5.2-5.4 in pI point on the autoradiogram. The spot for transferrin was shifted to the more basic side by treatment with monensin or tunicamycin for 3 hours. The spot for transferrin was also shifted to the more basic side by treatment with acetaldehyde for 3 hours or by additional treatment for 3 hours following pre-treatment for 6 hours with ethanol. However, the shift of transferrin was not observed with treatment with ethanol for 3 hours. These results suggest that glycosylation of secretory glycoproteins at the Golgi apparatus is inhibited by acetaldehyde, but not by ethanol itself, and that this inhibition may play an important role in the development of alcoholic liver disease.
