Stereo-Structures of Reaction Products of dl-1,3-Dimethylthymine Epoxide with Amines and L-Amino Acid Ethyl Esters(Organic,Chemical)

• Reaction of dl-1,3-dimethyIthymine epoxide (1) with amines (EtNH_2, tryptamine, β-Ala-OEt, benzylamine, Gly-OEt, morpholine, neopentylamine, and aniline) afforded cis and trans adducts (2 and 3). Reaction of 1 with L-amino acid derivatives (Pro-OEt, Met-OEt, Phe-OEt, and Trp-OEt) afforded four optically active diastereomers (4-7). The stereo-structures of the products were elucidated by X-ray analysis and by isomerization of the trans adducts to the cis adducts by using boron trifluoride etherate. A mechanism for the isomerization is proposed.
