Mucosal Protection by Shokyo, a Crude Drug, against Ethanol-induced Gastric Damage in Rat

The effects of Rikkunshi-to and each of its component crude drug against ethanolinduced gastric mucosal damage were compared in rats. Previous administration of Rikkunshi-to, Shokyo, Kanzo, Sojutu, Hange or Bukuryo (500mg/kg, p.o.) significantly prevented the gastric damage induced by 70% ethanol (p< 0.01 vs. control), and the most potent effect was observed with Shokyo (Zingiberis Rhizoma). Shokyo achieved the preventive effect against the ethanol-induced gastric damage in a dose-dependent manner, and considerable effect for the gastric protection was noted at 5mg/kg body weight. The antiulcerogenic effect of Shokyo was also indicated against hydrochloric acid-induced gastric damage but was not found against acidified acetylsalicylic acid or restraint and water immersion stress-induced injury. The protective effect of Shokyo against ethanol injury was not affected by indomethacin (5mg/kg) or N-ethylmaleimide (10mg/kg). The effect of Shokyo also remained in capsaicin-sensitive afferent neuron ablated rats. Shokyo (500mg/kg, p.o.) significantly prevented the decrease in mucin content of the corpus mucosa in ethanol-induced gastric injury. These results indicate that the protective action of Shokyo was mediated by neither generation of endogenous prostaglandin nor sulfhydryl compounds and might be independent from the capsaicin-sensitive afferent neurons. Mucus might be concerned with the preventive effects by Shokyo.
