Effects of 2-[(2-Dimethylaminobenzyl) Sulfinyl] Benzimidazole, the Proton-Pump Inhibitor, on Rat Gastric Mucins with HC1-Ethanol-Induced Gastric Damage

Examination was made of the correlation between the anti-ulcer effects of 2-[(2-dimethylaminobenzyl) sulfinyl] benzimidazole, the proton pump inhibitor, and gastric mucus glycoproteins (mucins). Considerable decrease in corpus mucins (46%) and antral mucins (51%) was observed in HC1-ethanol treated rats and HC1-ethanol reduced the amounts of fucose and galactose of corpus mucin. 2-[(2-dimethylaminobenzyl) sulfinyl] benzimidazole prevented macroscopical mucosal damage induced by HC1-ethanol. A single administration of this drug also caused reduction in mucosal mucins, though no mucosal damage or change in the carbohydrate component of mucins could be detected. In pylorus-ligated rats, this drug significantly inhibited gastric acid secretion by more than 90%. The reagent did not affect total mucin output in gastric juice, but secreted 1.5 times more concentrated mucin than that noted in the control group. The amounts of mucins in the gel layer increased 1.5 times as much with this reagent. This drug not only inhibits acid secretion but also promotes mucin secretion and increases mucus gel thickness and this may explain the cytoprotect of this drug against necrotizing agents.
