Effects of Acid Secretagogues or HC1 on Rat Gastric Mucins

The present study was carried out to compare the effect of endogenous and exogenous HC1 on gastric damage and mucus quality in rats. Rats were given histamine in doses from 0.08mg/kg to 80mg/kg per body weight intraperitoneally, tetragastrin 0.4〜120mg/kg subcutaneously, or 1 ml of 0.001〜0.6 N HC1 orally. The animals were examined one hour after administration of these reagents. Mucosal surface pH decreased in a dose-dependent manner for histamine, tetragastrin and HC1 except 0.6 N HC1. The high concentration of HC1 induced severe hemorrhagic erosions. Low dose histamine (0.8mg/kg) and tetragastrin (12mg/kg) increased corpus mucin, but HC1 did not exhibit this tendency. This increase in corpus mucin, however, was not inhibited by indomethacin. Low pH (<3) on gastric surfaces decreased gastric mucins, especially with exogenous HC1 and histamine. Hemorrhagic erosions likely result from the presence of these two conditions; the decrease of mucosal mucin and the decrease of mucosal surface pH. Macroscopic mucosal damage was produced by HC1 (0.35〜0.6 N) or histamine (8〜80mg/kg), but no lesion was induced by tetragastrin.
