Hyperglycemia-induced Alteration in Proteomes of the Rat Renal Cortex

We examined the effects of hyperglycemia on the proteomes of rat renal cortex in two types of diabetes mellitus, i. e., one was spontaneously developed type 2 diabetes mellitus in Otsuka-Long-Evans-Tokushima Fatty (OLETF) rats, and the other streptozotocininduced diabetes mellitus in Long-Evans-Tokushima-Otsuka (LETO) rats. Protein expressions were analyzed by two-dimensional electrophoresis (2-DE). Proteins separated by agarose 2-DE were identified by mass spectrometry (MS) and tandem mass spectrometry (MS/MS). In the renal cortices of diabetic rats, six proteins (pyruvate carboxylase, GRP78, argininosuccinate synthase, elongation factor-1 alpha, ubiquinol-cytochrome C reductase complex core protein 2, glutamate-oxaloacetic-transaminase) were up-regulated, and alpha 2u-globulin (A2u) was down-regulated. Among these proteins, A2u showed the greatest change in the diabetic rats. Immunohistochemical staining of renal cortices showed that A2u expression in the renal cortices was significantly lower in the glomeruli of hyperglycemic rats than in normoglycemic ones. We conclude that hyperglycemia plays an important role in both up-and down-regulation of proteins in the rat renal cortex.
