コレステロールとコール酸ナトリウム食給餌ラットにおける軽度の肝線維症(病理学)

• これまで高コレステロール血症に関する研究の大多数は, 粥状硬化症や冠動脈性心疾患に関して高コレステロール食の影響について行われていた.しかし, 肝に対するコレステロールの毒性や, 高コレステロール食摂取と肝線維症の関係については, 明確にあるいは病理組織学的に検索されていない.雄Wistarラットが1.0%コレステロールと0.3%コール酸ナトリウムを加えた飼料を12週間給餌された.ラットは安楽殺され, 血液生化学, 通常の顕微鏡ならびに免疫組織化学的に検査された.この飼料の摂取で, ラットのアスパラギン酸アミノトランスフェラーゼ, アラニンアミノトランスフェラーゼ, 乳酸脱水素酵素, 総コレステロール値は, 3週以降実験期間中上昇していた.顕微鏡観察では, 肝の壊死, マクロファージの浸潤, 脂肪化が実験期間中著しく増加した.肝の線維増生と筋線維芽細胞の増殖は, 9及び12週でみられた.マスト細胞の出現は, 肝傷害の程度に比例していた.これらの所見は, 肝線維症は高コレステロール食によって誘導されうること, しかもそれが炎症環境における何種類かの細胞(すなわちマクロファージ, 筋線維芽細胞, マスト細胞)間の相互作用の結果であることを示唆している.高コレステロール血症は, 粥状硬化症や冠動脈性心疾患と同様に, 肝線維症に対する危険因子であると考えられる.
