Expression of GAP-43 mRNA in the myenteric nerves following transection in the muscle coat of the rat small intestine

• We investigated the expression of Growth Associated Protein (GAP)-43 on the regrowing myenteric nerves in the rat small intestine by immunohistochemistry and in situ hybridization. Following myotomy, the severed area was examined at various intervals by 7 days after the operation. Just behind the operation, GAP-43-immunoreactivity rapidly declined in the myenteric nerves both on the oral and anal severed stumps. At approximately 12h after the operation, accumulation of GAP-43-immunoreactivity was seen in nerve cell bodies proximal to the cut edge. On postoperative day 2, marked increase in the intensity of the immunoreaction was found in the transected nerves extruding fine fibers. On day 7 after the operation, a great number of GAP-43-immunoreactive fiber bundles was detected in the severed area. The distribution of signals for GAP-43 mRNA were primarily demonstrated in regenerating neurites as well as in neuropils and neuronal somata under normal conditions. Signals for GAP-43 mRNA were mostly tracked by the immunolocalization of the protein in the course of the nerve regeneration. In conclusion, our findings suggest that GAP-43 may play a significant role in myenteric nerve regeneration and plasticity by means of both the transport of GAP-43 transcripts to the neurite extensions and the synthesis of the protein in focal area.
