PURIFICATION AND PROPERTIES OF A HUMAN PLACENTAL POST-PROLINE ENDOPEPTIDASE

Post-proline endopeptidaseはprolineを含むpeptideの-Pro-X結合を分解する酵素である.妊娠時に重要な生理作用を示すTR-H,oxytocin,angiotensinには全てprolineが含まれ,妊娠時に著増するoxytocinaseはこれらをN末端から順次加水分解するもののproline部位にて反応は停止する.よつて,-Pro-X結合分解酵素の重要性が示唆され,ヒト胎盤に本酵素の証明及び精製を行い,その性質を明らかにする事を試みた.ヒト正常妊娠胎盤を材料とし,酵素活性はSuc-Gly-Pro-MCAを基質としKatoらの方法で測定した.胎盤から硫安分画,phenyl-Sepharose,DE-52,Z-Gly-Pro-Leu-Gly-Pro-AH-Sepharose,Bio-Gel A-0.5mの各chromatographyにより精製し,精製倍率3390借,比活性1.76μmolAMC/mg/min,活性回収率14.9%であつた.精製酵素はpolyacrylamide-gel電気泳動及びSDS-gel電気泳動上,蛋白質としてほぼ単一bandを示した.至適pHは6.7,等電点は4.8,Suc-Gly-Pro-MCAに対するK_mは1.0mM,V_<max>は5.3μmol AMC/mg/minであつた.分子量はgel filtrationにて140000,SDS-gel電気泳動にて67000と推定された.本酵素はSH基阻害剤にて強く活性抑制されthiol proteaseである事が示唆された.ヒト胎盤では初めて本酵素を証明並びに精製し,酵素化学的性質を明らかにした.prolineを含む各種peptide hormoneの代謝に胎盤中の本酵素が重要な役割を示す事が考えられる.
