DISSOCIATION OF hCG-RECEPTOR COMPLEX AND DETECTION OF hCG RECEPTORS IN HUMAN CHORIONIC TISSUES

我々は, hCGレセプターbinding実験のモデルとしてラット睾丸を用いてhCGレセプター結合物の解離方法とそのラジオレセプターbindingアッセイを確立した. hCGレセプター結合物はdithiothreitol, neuraminidaseとlow pH buffer (3.3)の処理により解離し, 解離率は dithiothreitolで42.8%, neuraminidaseで35.2%とpH (3.3) bufferで68.7%であつた. 処理後のレセプターは [^<125>I]hCGとrebindingができ, 結合常数は1.798〜3.496%/pMであつた. 高濃度のNaClとMgCl_2では解離できず, Triton X-100と高濃度のMnCl_2処理したものはrebinding activityを失つた. 以上の方法を利用し, 11例の胎盤(8週, 11週, 16週, 24週各1例とterm 7例)のhCGレセプターを調べた. それぞれの処理により絨毛組織への [^<125>I]hCG bindingは139〜226%増えたが, このbindingはcold hCGにより抑制されず, Scatchard分析はbindingの特異性を証明できなかつた. 絨毛癌細胞株BeWoにおける [^<125>I]hCG binding実験でも同様の結果を得た. 以上より胎盤絨毛にはhCGレセプターは存在しないと考えられる.
