ESTABLISHMENT OF HYBRIDOMAS SECRETING MONOCLONAL ANTIBODIES TO PLACENTAL ALKALINE PHOSPHATASE AND DEVELOPMENT OF AN ENZYME IMMUNOASSAY FOR ITS DETERMINATION

胎盤性アルカリフォスファターゼ(PLAP)に対するマウスモノクローナル抗体(MoAb)を産生する7種類のハイブリドーマを樹立し, それぞれ(1) 7C6, (2) 6G10, (3) 5B9, (4) 6D5, (5) 6B5,(6) 11G6, (7)3E10, と命名し, radioimmuno-assayによりそれらの抗体の性状を調べた. 各抗体の小腸性アルカリフォスファターゼとの交差性は, (1) 0.04, (2) 0.2, (3) 1.4 (4) 1.8, (5) 0, (6) 4.0, (7) 6.2 (%),であった. 一方, 肝性アルカリフォスファターゼとは2,000IU/1まで交差性を示さなかった. ついで, それらのうちでPLAPに対する特異性と親和性において最も優れた抗体である7C6を用いてenzyme immunoassay (EIA)を作成し, 各種婦人科疾患患者血清中のPLAP値を検討した. 健常女性婦人に対するcutoff値を0.1IU/1と設定すると, 婦人科良性疾患では6.4%, 子宮頚癌では21.5%, 子宮内膜癌では36.4%, 卵巣悪性腫瘍では39.5%の陽性率が得られた. CA125に比べ, 悪性疾患に対して高い特異性が認められた. 子宮内膜癌の中では, 高分化型腺癌に高い陽性率(47.4%)が得られ, また卵巣悪性腫瘍の中では, 漿液性嚢胞腺癌(41.4%), 類内膜癌(50%), 未分化胚細胞腫(100%)およびKrukenberg腫瘍(50%)において陽性率が高かった.
