Characteristic Changes of Reducible Collagen Cross-links to the Hard Tissue Pattern in Turkey Tendon upon Calcification : Nature of Calcifiable Collagen

Calcifying turkey tendon is known as a good model of collagen mineralization due to the adjacent presence of discrete uncalciried and calcified tissues. Collagenous tissues were isolated from both adjacent uncalcified and calcified portions of turkey tendon, and were processed using the same method of deminera1ization. Reducible cross-links of collagens of both portions were analyzed on ionexchange HPLC after sodium borohydride reduction and hydrolysis. The cross-link pattern of col1agen from the uncalciried portion was similar to that of skin, being rich in hydroxylysinonorleucine and histidinohydroxymerodesmosine. On the other hand, the pattern on the calcified portion of collagen was simllar to that of bone and dentin, being rich in dihydroxylysinonorleucine and scarce in histidinohydroxymerodcsmosine. The results indicate that during the calcification of tendon collagen, additional synthesis of new collagen and/or some rearrangement of cross-1inking,. which led the preferential formation of dehydro-dihydroxylysinonorleucine, occurred.
