Chemotactic Response of Periodontal Ligament Cells toward rhBMP-2

Periodontal ligament cells(PDL cells)are responsible for the functions and maintenance of periodontal tissue. However, little is known about the regulatory mechanism for PDL cell activity. The aim of this study is to confirm whether PDL cells possess chemotactic activity toward recombinant human bone morphogenetic protein 2(rhBMP-2). Fresh, healthy PDL cells were obtained from the first and second premolars of orthodontic patients. It was found that chemotactic activity of rhBMP-2 for PDL cells started at 1-10 ng/ml of rhBMP-2, gradually increased in a dose-dependent manner, reached the highest value at 5 μg/ml, and then decreased at 10 μg/ml. On the other hand, PDL cells began to react with basic faibroblast growth factor(bFGF)at 10^<-2>ng/ml, reached the maximum reaction at 10 ng/ml. The high reactions continued until the concentration of 10^3 ng/ml and decreased thereafter. Overall reactions of PDL cells toward bFGF were much higher than those toward rhBMP-2. In contrast to the PDL cells, chemotactic response of osteoblast-like cells MC3T3-E1(E1 cells)toward rhBMP-2 and bFGF showed both in a typical bell-shaped manner, were attained maximal concentration of 100 and 10 ng/ml, respectively. The results indicate that PDL cells and E1 cells have their feasible cytokines, toward which they are induced chemotaxis and also have optimal concentrations of the cytokines for this reaction. The specificity in chemotactic reactions of these two cytokines for PDL cells and osteogenic E1 cells is important to note in therapeutic applications for periodontal regeneration.
