Effects of mechanical stress on the eryression of type XII collagen mRNA in human periodontal ligament cells

The aim of this study is to analyze the effect of mechanical stress on the formation and maintenance of the periodontal ligament(PDL). Type XII collagen was chosen as a characteristic marker of extracellular matrix, together with three other noncollagenous proteins, osteopontin, osteonectin and osteocalcin. Human PDL cells were cultured and subjected to intermittent tensile stress using a Flexercell Strain Unit^<TM>(1/20Hrz, 18% elongation for 1, 3, 5 or 7days). By the RT-PCR method with a LightCycler^<TM> mRNAs of these proteins were quaentitatively analyzed. It was found that the mRNA expression of collagen XII, osteocalcin and osteonectin was increased by the mechanical stress in a time-dependent manner, wheres no significant change of mRNA of osteopontin did not observed. The results suggested that intermittent stress positively contributes to the formation and maturation of PDL cells to form the PDL.
