Laser perforated collagen membrane : pore size-dependent bone induction as a new BMP carrier

Bone morphogenetic protein(BMP)requires a certain carrier to induce bone when implanted into animal tissues such as muscle, skin and periodontal tissues. More than ten different BMP-carriers have been developed to meet different clinical needs. To develop a membranous carrier that is biodegradable, we newly prepared laser perforated collagen membranes(LPM)(35 mm in thickness, non-cross-linked pepsin-treated collagen)with different pore sizes ranging from 0.1 to 1.0 mm in diameter. Geometrical similarity was used to keep a constant ratio of the perforated to the nonperforated areas of the membrane. We compared their efficaies for bone formation in BMP-induced ectopic osteogenesis. The membranes were impregnated with rhBMP-2, implanted subcutaneously into rats and analyzed after being extracted at 1-4 weeks. It was found that all of the LPM showed higher bone-inducing efficacy than the non-perforated collagen membrane as shown by□@histological observations, calcium contents and the alkaline phosphatase level. □@LPM with pore sizes larger than 0.5 mm showed higher amounts of bone than the non-perforated membranes, with the 3.3 times the calcium content(at 3 weeks). These results suggested that the retention of the cytokine and the anchorage of the recruited mesenchymal cells on LPM, particularly with pore sizes larger than 0.5 mm, were higher than those of non-perforated membranes. The LPM is applicable for tissue engineering in the dental field.
