Effects of LPS and Protease from Porphyromonas gingivalis on Cultured Human Gingival Fibroblasts

Lipopolysaccharides (LPS) and proteases produced by Porphyromonas gingivalis (P.gingivalis) are both significant pathogenic factors of periodontitis. In this study, we determined how LPS and the protease produced by P. gingivalis affect cultured human gingival fibroblasts (HGF). The protease was isolated from the culture supernatant of P. gingivalis. LPS was prepared from P. gingivalis 381 by the hot phenol-water extraction method. Normal human gingival tissue specimens were obtained from gingival biopsies and the tissue was cultured. HGF were stimulated with P. gingivalis LPS, the protease, or both for up to 24 h. The rates of cell growth in the cultures was measured by MTT assay using an optical density reader. The fibronectin levels on the cell surface of HGF were measured by enzyme-linked immuno sorbent assay (ELISA). The production of interleukin-1 β (IL-β) was also measured by ELISA. The growth rate of HGF was increased by P. gingivalis LPS, but decreased by P. gingivalis protease. Likewise, the level of fibronectin in HGF was increased by LPS, but decreased by P. gingivalis protease. Although the production of IL-1β by HGF increased by the addition of LPS alone, the production decreased when both LPS and protease were added. These findings suggest that P. gingivalis LPS activates HGF, and P. gingivalis protease destroys the cell structure, specifically the cell-matrix of HGF.
