BMP-Induced Osteogenesis with Two Geometrically Different Biodegradable Carbonate Apatites

The purpose of this study was to establish a tool for local bone reconstruction by using BMP combined with biodegradable carbonate apatite(CAP), and to find a feasible geometry for biodegradability of the BMP carrier. Two types of CAP particles(0.3-0.5 mm in average size)with different surface geometries were prepared by a high-pressure method. One type was pressed and equipped with high specific density(P-CAP), and the other type was non-pressed with lower specific density(NP-CAP). The P-CAP, NP-CAP and the porous particles of hydroxyapatite(PPHAP)were combined with 5 μg of rhBMP-2(a gift from Yamanouchi Co.)and implanted subcutaneously into the backs of rats. The implants were analyzed at 1, 2, 3 and 4 weeks, and 1, 2, 4, 6, 8 months for histology, osteocalcin contents and alkaline phosphatase(ALP)activity. Direct osteogenesis without detectable chondrogenesis was observed in all three carriers with BMP. The NP-CAP showed highest ALP(P<0.05)at 2 weeks. The PPHAP and NP-CAP showed much higher osteocalcin contents(P<0.02)than those of P-CAP at 4 weeks. The results indicated that bone-inducing efficacy was higher in the rough-surfaced NP-CAP than in the smooth-sur-faced P-CAP. In addition, histometrical analysis showed that the volume of the NP-CAP im-plants significantly decreased after 6 months(P<0.02), and 8 months(P<0.01)compared with those of 2-month implants. The results of histometry verified the biodegradability of the NP-CAP when implanted with BMP.
