Ultrastructural changes in serial liver biopsy specimens from chimpanzees inoculated with the agent of non-A, non-B hepatitis and their reactivity with the 48-1 antibody.

Characteristic ultrastructural changes in hepatocytes; tubular structures, microtubular aggregates, and sponge-like inclusion, have previously been reported for experimental non-A, nonB hepatitis in chimpanzees. To further investigate the relationship between appearance of these structures and non-A, non-B hepatitis, electron microscopic examinations were carried out on serial liver biopsy specimens from chimpanzees acutely, chronically, or asymptomatically infected with the agent of non-A, non-B hepatitis.<BR>The structures were observed in the biopsies obtained during preacute, acute, and chronic phase of hepatitis, but not in those taken during convalescent phase or prior to inoculation.<BR>The structures were also detected in an asymptomatic case; a chimpanzee who did not develope the serum transaminase elevation was positive as well. Thus it seemed that appearance of the structures might be associated with the viral infection rather than cell destruction.<BR>In addition, the reactivity of the structures with the monoclonal antibody (48-1) previously shown to be related to non-A, non-B hepatitis (Shimizu, et al. Proc. Natl. Acad. Sci. USA 82, 1985) was examined by immunoperoxidase electron microscopy.<BR>The result showed that the 48-1 antibody reacted with the microtubular aggregates, but not with tubular structures or sponge-like inclusions.
