Functional analysis of the pre-C region of DHBV DNA: A transfection study of the pre-C frameshift-mutated DNA.

To analyze the function of the pre-C region of DHBV DNA, we introduced a frameshift mutation into the Acc I site in this region, and transfected this mutant DNA into the liver of 1-day-old ducklings. The ducklings transfected with mutant DNA developed DHBV infection, and this mutant virus was also infectious to susceptible ducklings. The direct sequencing confirmed the presence of pre-C mutation in the progeny genomes. By Western blotting, no DHBeAg was detectable in serum of ducks infected with pre-C mutant virus. The histological examination showed more active histological changes in wild-type than in pre-C mutant. These results suggest the pre-C region is not essential for viral replication and infection, but required for the systhesis and secretion of DHBeAg, and it has become possible to study the pathogenicity of the DHBeAg producing/nonproducing virus in liver.
